寄生虫に対する免疫応答(II型免疫応答)の仕組み

免疫学の教科書を読んでいると、病原体が細菌か、ウイルスか、寄生虫かで対応する免疫応答が異なるようです。

細菌の場合はマクロファージなどの貪食細胞が活躍します。また、樹状細胞がその細菌を貪食して細菌由来のタンパク質を分解しペプチドの形で抗原提示をT細胞に対して行い、たまたまその抗原にピッタリのT細胞抗原受容体をもっていたナイーブヘルパーT細胞が活性かされて活性化ヘルパーT細胞(Th1)になります。同じ抗原たんぱく質を認識するナイーブB細胞と出会ったヘルパーT細胞はナイーブB細胞を活性化し、抗体を産生するプラズマ細胞へと変化させます。抗体が細菌に結合すると(オプソニン化)、マクロファージによる貪食能が増進します。また、ヘルパーT細胞はマクロファージにも働きかけて、貪食能を増進させます。

侵入者がウイルスだった場合には、細胞傷害性T細胞が活躍し、ウイルスに感染した細胞を直接死に至らしめます。

さて、侵入者が寄生虫だった場合はどうなるでしょう。この場合に発動する免疫反応は、花粉に対する反応と同じでいわゆるII型免疫応答と呼ばれるものです。II型というのは、ヘルパーT細胞の方がTh2という2型であり、また、自然リンパ球II型からもTh2と同様のサイトカインが放出されて、これらのサイトカインが活躍するからです。

英語の論文や教科書を読むと寄生虫はhelminthsと書かれています。これは「蠕虫」と訳されていて、くねくねとした動き(蠕動)により動く虫だからこういう名前になっているようです。「寄生虫」は、単細胞生物である「原虫Protozoa」と多細胞生物である「蠕虫Helminths」とに大きく2分されます。原虫に属するお馴染みの名前のもととしては、赤痢アメーバ、トリパノソーマ、膣トリコモナス、マラリア、トキソプラズマといったものたちがいます。蠕虫はさらに、線虫類Nematoda、吸虫類Trematoda、条虫類Cestoideaなどに分かれます。線虫類は、回虫(回虫、アニサキス)、蟯虫、遠景線虫などの種類があります。吸虫類には日本住血吸虫などがいます。

樹状細胞は寄生虫をどのように認識するのか

細菌の場合は樹状細胞が細菌を貪食して細胞内にとりこんでプロテアーゼで分解してペプチドにして抗原提示するというシナリオはなるほどと思えるのですが、寄生虫のように大きなもの(体長数mm~数cm)を分解してタンパク質を取り込むというのが想像できません。教科書を読んでもレビュー論文などを読んでも、そのあたりを明確に書いているものに出会えていません。

上の説明だと樹状細胞が認識するのは蠕虫(ぜんちゅう)の卵のようです。卵なら単細胞なので貪食できそうですね。

花粉アレルギーもそうですが、寄生虫に対する免疫応答はなぜTh2/Th1バランスがTh2に傾くのでしょうか。上の論文はTLRの何番が何の物質の受容体になっているということを細かく述べていましたが、実験では蠕虫の抽出物を用いているので生体の状態の蠕虫が樹状細胞にどのように認識されるのかのイメージがまだ湧きません。表面にペタッとつくだけでも、膜表面のタンパク質を捉えることができるのでしょうか。蠕虫に樹状細胞が引っ付いているような状態を観察した写真というものがネット上を探しても全然見つかりません。

上の説明だと蠕虫が分泌するタンパク質に樹状細胞が反応するようです。ただこれだと、分泌されたタンパク質に対する抗体ができてもその抗体は成体には反応しなくて、成体の攻撃にはならないのではないでしょうか。まだしっくりきません。

上の図でも別に寸法を正しく描いているわけでもないので、もやっとした説明でしかないように思います。

B細胞は寄生虫をどのように認識するのか

B細胞が抗体を産生するためにはヘルパーT細胞のヘルプが必要ですが、そのためにはB細胞も寄生虫のタンパク質を貪食してペプチドにまで分解したものを抗原提示する必要があります。寄生虫のような大きなものをどうやって抗原として取り込めるのでしょうか。
